BioTek 50 TS Microplate Washer

The BioTek 50 TS from Agilent is a compact, programmable automated plate washer validated for use with Mabtech's EYRAplex, ELISpot, FluoroSpot, and ELISA assays. Use our own validated wash programs for each assay and move from washing to results faster. The 8-well manifold washes eight wells at a time, so an entire 96-well plate is washed in under 5 minutes.

Two versions are available. The magnetic version handles magnetic bead-based EYRAplex assays as well as ELISA, ELISpot, and FluoroSpot. The non-magnetic version is for ELISA, ELISpot, and FluoroSpot.

  • Validated for EYRAplex, ELISpot, FluoroSpot, and ELISA
  • Pre-optimized Mabtech wash protocols, with no method development needed
  • Magnetic version includes a flat magnet for EYRAplex and other magnetic bead-based assays
  • Efficient buffer removal without bead loss in magnetic washing
  • One washer for your entire Mabtech workflow, magnetic and non-magnetic assays
  • Fully programmable volume, soak time, number of cycles, and aspiration height
  • Eight-well manifold for 96-well plates, washing 8 wells at a time

Ease of use

The built-in touch screen and simple menu design allow users to create and save custom protocols or choose from predefined protocols with commonly used wash parameters. The intuitive interface and menu-driven software make your assays just that much easier. The user manual includes all the information you need to perform and program washes as well as how to run maintenance routines for optimal performance.

Applications

The Agilent BioTek 50 TS Magnetic Microplate Washer has been validated for all Mabtech assays. We've optimized each wash program for you so you can get right to your assay and results. If you need to customize for other assays, you can adjust aspirate height, fluid delivery times, and more in just a few clicks.

Dedicated wash programs for ELISA, ELISpot, FluoroSpot, and EYRAplex, validated for each assay, ensure proper buffer and wash dispensing and removal, improving assay robustness.

Automated maintenance routines

Routine maintenance ensures your washer is ready when you need it, delivering continuous and reliable performance. Predefined, automated maintenance routines include daily and even overnight protocols to keep the fluid path clean and prevent buildup of salt, protein, or other material that could block the manifold tubes, leading to inadequate washing. Everything is laid out in the included user manual.

Pre-programmed, automated mainentance routines clean the washer fluidics tubing and probes for dispensing and aspiration to prevent build up of salts from wash buffers and assay diluents. Run regularly as outlined in the washer manual for smooth and long-lasting performance.

Find out which analyte combinations we have evaluated in T cell FluoroSpot and which combinations are affected by capture effects or not.

What is a capture effect?

When capture antibodies with different specificities are coated together, the capture of one cytokine may affect the secretion of other cytokines. This is usually more pronounced when studying T cell responses with polyclonal stimuli compared to antigen-specific responses. Capture effects are seldom a problem and can often be counteracted by the addition of an anti-CD28 antibody.

Depiction of capture effects observed in FluoroSpot

(1) IL-2 secreted by the activated T cell is captured by coated anti-IL-2 capture antibodies. (2) As a result, IL-2-stimulation of the T cell itself (autocrine stimulation) as well as nearby T cells (paracrine stimulation) is impaired, ultimately leading to (3) decreased IFN-γ secretion.

Co-stimulation with anti-CD28

Anti-CD28 mAb provides a co-stimulatory signal to antigen-specific responses by binding to CD28 on T cells. The addition of an anti-CD28 mAb to the cell culture enhances antigen-specific responses and can counteract capture effects. For example, the presence of IL-2 capture antibodies may result in reduced activation of T cells, as capturing of IL-2 decreases the amount of available IL-2 and thereby dampens the IL-2-mediated stimulation of T cells. The addition of anti-CD28 mAb restores IFN-γ responses (depicted in the below images). Further optimization may be necessary, depending on the cells and stimuli used. Too high concentrations of the anti-CD28 mAb may result in an elevation of non-specific cytokine secretion.

Overcoming capture effects using anti-CD28 anitbodies in FluoroSpot

(1) An anti-CD28 antibody can be added to provide a co-stimulatory signal that can restore (2) e.g. IFN-γ responses.

How to investigate capture effects

Our FluoroSpot Plus kits are evaluated for capture effects, and in studies of T cell responses we recommend co-stimulation with anti-CD28. With FluoroSpot Flex, it is possible to combine and build your own kit. For guidance look at our analyte combination table (above). Capture effects can be investigated by quantifying spot numbers in wells coated with single capture antibodies and compared to wells coated with a mixture of the capture antibodies. The compensatory effect of the anti-CD28 mAb may be assessed by comparing cells cultured with and without the anti-CD28 mAb.
