Plate Preparation protocol of using Pre-coated ELISpot clear plate

    Dear Mabtech representative,

    I'd like to ask question about the protocol of using Pre-coated ELISpot clear plate (code: 3420-3PT) specifically a the plate preparation step. The Guidlines stated to remove the plate from the sealed package and wash 4 times with sterile PBS (200 uL/well). Since I don't have the sterile PBS, is it okay to use sterile RPMI instead of sterile PBS? And what could be the consequences of using the RPMI as the substitute? And if using RPMI is not a good alternative, do you have any recommend of solution that can be used to prepare the plate?

    Using sterile RPMI will work well. Doable.

    The idea of the first wash is simply to condition the plate. As long as the liquid is sterile and dont have weird components or strange pH you are fine.

    Thank you for the reply. It is very helpful. I want to make sure, If I use sterile water with pH 7, is that also okay?

    No please do not use water.

    Water can have different levels of pH and can negatively affect the capture mab.
