Intended use

This ELISA Flex kit is intended for the quantification of reduced and oxidized forms of human Thioredoxin-1 (Trx1) in solution. For research use only. Not for use in diagnostic procedures.

Serum/Plasma samples

Analysis of serum/plasma with this kit requires the use of Assay buffer. The Assay buffer blocks heterophilic antibodies from cross-linking the assay antibodies, thereby preventing false-positive read-outs. Heterophilic antibodies are commonly found in human serum/plasma. The Assay buffer has been validated using from healthy human blood donors. It should be used for dilution of standard, samples, and detection antibody.For quantification of Trx1 in blood, the use of plasma is recommended since serum contains high levels of Trx1 released from platelets during sample preparation. To minimize the platelet content in the plasma sample, two sequential centrifugations of plasma at 10,000 x g for 10 minutes are recommended.

Thioredoxin-1

Analyte description

Thioredoxin-1 (Trx1) is an oxidoreductase protein and thus may act as an anti-oxidant by facilitating the reduction of cysteine residues of other proteins. Besides keeping the intracellular milieu in a reduced state and being involved in the defence against oxidative stress, thioredoxin-1 specifically regulates several transcription factors and receptors. Increased plasma levels of thioredoxin-1 has been associated with several psychiatric disorders.
