Intended use

This monoclonal antibody enables specific detection of the influenza A nucleoprotein (NP) in FociSpot immunostaining of virus infected cells. The antibody can also be used in ELISA.

FociSpot immunostaining of influenza A nucleoprotein in focus forming assays. Antibody functionality in FociSpot was confirmed using influenza A infected MDCK cells.

Performance

Detection of influenza A nucleoprotein in indirect ELISA. An ELISA plate was coated with recombinant influenza A nucleoprotein. A dilution of anti-influenza A (NP) mAb 19C10 (biotinylated) was added and the plate was developed using Streptavidin-HRP and TMB substrate.

FociSpot immunostaining of influenza A nucleoprotein in infected MDCK cells. MDCK cells were infected with influenza A virus (titrated, virus dilutions 2,000x, 4,000x, and 8,000x are shown). After fixation and permeabilization, foci were detected using anti-influenza A (NP) mAb 19C10, followed by Streptavidin-ALP and BCIP/NBT-plus substrate. Foci were counted using the Apex software in Mabtech IRIS 2.

Analyte description	The Nucleoprotein (NP) is a structural protein and encapsidates the negative strand RNA of influenza A into ribonucleoproteins (RNP). NP plays a role in replication, transcription, RNA packaging, and nuclear trafficking. The NP does not only interact with the RNA, but also the viral polymerase, the M protein, and several host factors.
