Bovine PBMC were incubated overnight in the presence (40,000 cells/well) or absence (200,000 cells/well) of PHA and the number of cells secreting IL-17A was analyzed by ELISpot.
Sheep PBMC (100,000 cells/well) were incubated overnight in the presence or absence of ConA and the number of cells secreting IL-17A was analyzed by ELISpot.
Goat PBMC (100,000 cells/well) were incubated overnight in the absence of PHA and the number of cells secreting IL-17A was analyzed by ELISpot.

This monoclonal antibody enables specific detection of bovine, sheep, goat, and porcine IL-17A in immunoassays such as ELISpot, FluoroSpot, and ELISA.

Recommendation

MT49A7 is recommended as capture mAb in ELISpot, FluoroSpot, and ELISA in combination with detection mAb MT51B8 (product code 3109-6) for detection of IL-17A from cow, sheep, and goat. For detection of IL-17A from pig, MT49A7 should be used in combination with detection mAb MTP853 (product code 3020-6).

Analyte description

Interleukin 17A (IL-17A) is a potent proinflammatory cytokine produced by activated Th17 (T helper 17) cells and certain cells belonging to the innate immune system. In mice, IL-17 has also been shown to be produced by activated CD8 T cells and γδ T cells. Th17 cells play an important role in autoimmune diseases and protection against bacteria and fungi. IL-17A acts on a broad range of cell types to induce the expression of cytokines, chemokines, and metalloproteinases. As a result, secretion of IL-17A promotes inflammatory responses, which leads to the recruitment of neutrophils, enhancement of antibody production, and activation of T cells. Increased expression of IL-17A is seen in autoimmune diseases such as multiple sclerosis and rheumatoid arthritis. It is also associated with asthma, psoriasis, cancer, and transplant rejection.
