Intended use

This ELISA Pro kit is intended for the quantification of human apoB in serum, plasma, and cell culture supernatants. For research use only. Not for use in diagnostic procedures.

Serum/Plasma samples

The Apo ELISA buffer included in this kit blocks heterophilic antibodies from cross-linking the assay antibodies, thereby preventing false-positive read-outs. Heterophilic antibodies are commonly found in human serum/plasma and can also be present in other species. The Apo ELISA buffer has been validated using from healthy human blood donors.

Apolipoprotein dilution
Apolipoproteins are present at high levels in serum and plasma. For ELISA quantification, such samples must therefore be diluted 5,000-200,000 times depending on the apolipoprotein analyzed.

Specificity

This kit is based on a matched pair of mAbs specific for human apoB. The assay is specific for apoB100 and does not recognize apoB48.

Cross-reactivity

The mAbs react with human apoB and cross-react with apoB from non-human primates (NHP). For more information please check the Cross-reactivity guide section below.

Analyte description

Apolipoprotein B (apoB) is the main protein component of low-density lipoprotein particles (LDL) and it is a large water-insoluble molecule of 512 kD (apoB-100) produced mainly in the liver and in the intestine (apoB-48). ApoB is regarded as a negative factor in cardiovascular disease. ApoB-100 is normally 98% of the apoB in human plasma and at a concentration of approximately 0.4-1.3 mg/ml.
