Product specifications

Intended use

This monoclonal antibody enables specific detection of apoA1 in immunoassays such as ELISA and Western blot.

Serum/Plasma samples

Recommendation

HDL 44 is recommended as detection mAb in ELISA in combination with capture mAb HDL 110 (product code 3710-3). The antibody is also suitable for immunoprecipitation and Western blot.

Specificity

Human apolipoprotein A1 (apoA1), purified or in the form of HDL. The presence of low amounts of detergent is necessary for binding. Tween 20, Triton X100 or NP40 can be used at a concentration of 0.01-0.5%.

Cross-reactivity

Cross-reacts with apoA1 from rhesus macaque and cynomolgus macaque. For information on reactivity with other non-human primates (NHP), please check the NHP cross-reactivity guide.

Purification

Purified from in vitro cultures by protein G affinity chromatography.

Biotinylation

Biotinylated through reaction with a N-hydroxysuccinimide ester of biotin.

Storage

Store product at 4-8°C or frozen at -20°C or below. Avoid repeated freezing/thawing.

Shelf lifeAt least 18 months from date of receipt.
Detection of human apoA1 with mAb HDL44 in Western blot. Human plasma (0.1 µl and 0.01 µl) in SDS sample buffer were denatured at 70˚C for 10 minutes and run on SDS-page / Western blot. ApoA1 was detected using 1 µg/ml HDL44 (biotinylated) followed by SA-ALP and BCIP/NBT-plus substrate.

The systems reactive with NHPs are either based on cross-reactive human kits or specifically developed monkey kits (NHP).

Cross-reactivity verification tests have been performed in ELISpot and/or ELISA by Mabtech and/or by others. Evaluations of assays with less solid evidence of cross-reactivity are shown as symbols within parentheses.

apoA1

Analyte description

Apolipoprotein A1 (apoA1) is the main protein component of high-density lipoprotein particles (HDL). ApoA1 (28 kD) is mainly produced in the liver and exported into circulation where it acquires lipids and becomes HDL. A high level of apoA1 is considered protective against atherosclerosis and cardiovascular disease. The normal concentration of apoA1 in human plasma is approximately 0.8-2.2 g/L.
