Intended use

This ELISA Pro kit is intended for the quantification of human apoE in serum, plasma, and cell culture supernatants. For research use only. Not for use in diagnostic procedures.

Serum/Plasma samples

The Apo ELISA buffer included in this kit blocks heterophilic antibodies from cross-linking the assay antibodies, thereby preventing false-positive read-outs. Heterophilic antibodies are commonly found in human serum/plasma and can also be present in other species. The Apo ELISA buffer has been validated using serum/plasma samples from healthy human blood donors.

Apolipoprotein dilution
Apolipoproteins are present at high levels in serum and plasma. For ELISA quantification, such samples must therefore be diluted 5,000-200,000 times depending on the apolipoprotein analyzed.

Specificity

This kit is based on a matched pair of mAbs specific for human apoE. The ELISA detects the three apoE isoforms apoE2, apoE3 and apoE4.

Cross-reactivity

The mAbs react with human apoE and cross-react with apoE from non-human primates (NHP). For more information please check the Cross-reactivity guide section below.

Shipping and Storage

Shipping

Shipped at ambient temperature.

Storage

Store detection mAb, Streptavidin-HRP, buffers/solutions, and substrate at 4-8 °C upon receipt. The ELISA standard should be stored at -20 °C. Plates may be kept at room temperature.

Shelf lifeAt least 12 months from date of receipt.
Standard range0.02-15 ng/ml
Sensitivity0.01 ng/ml

The lowest concentration that can be detected, but not necessarily quantified with precision and accuracy. This was determined by adding 2 standard deviations to the mean OD of background wells.

Mean standard curve from repeated runs of human apoE ELISA Pro kit.

The systems reactive with NHPs are either based on cross-reactive human kits or specifically developed monkey kits (NHP).

Cross-reactivity verification tests have been performed in ELISpot and/or ELISA by Mabtech and/or by others. Evaluations of assays with less solid evidence of cross-reactivity are shown as symbols within parentheses.

apoE

Analyte description

Apolipoprotein E (apoE) is a 34 kD glycoprotein involved in lipid transport, lipoprotein metabolism, and cellular signaling. It is present in plasma lipoprotein particles such as chylomicrons, VLDL, and HDL. ApoE is also present in large amounts at sites of neurological damage and appears to be involved in the recycling of apoptotic remnants and amyloidal aggregates. ApoE is mainly produced in the liver and by macrophages, but also by other cell types such as smooth muscle cells or neuronal cells. In humans, apoE exists in three isoforms, apoE2, apoE3, and apoE4. Of these, apoE4 is associated with Alzheimer’s disease. A normal concentration of apoE in human plasma is approximately 30-40 µg/ml. The concentration of ApoE in inbred mouse plasma can vary between 15-150 µg/ml.
