B-cell FluoroSpot kit for enumeration of cells secreting human IgG/IgA

The FluoroSpot assay is an inventive development of the ELISpot assay and uses fluorescent detection instead of colorimetric. This allows for simultaneous detection of several analytes. The human IgG/IgA B-cell FluoroSpot kit enables detection of IgG- and IgA-secreting cells in the same well.

The kit contains matched quantities of capture antibodies and fluorophore-conjugated detection antibodies, low fluorescent PVDF plates and a fluorescence enhancer. The polyclonal activators R848 and recombinant human IL-2 are included for pre-stimulation of memory B cells.

For analysis, we recommend the use of an automated FluoroSpot reader equipped with filters for FITC (excitation 490 nm/emission 510 nm) and Cy3 (excitation 550 nm/emission 570 nm). Filters should have high specificity to avoid bleed-through artifacts.


Analyte Information

IgA
IgA (Immunoglobulin A) represents only 10-15% of the total Ig molecules in serum but is the predominating Ig isotype in saliva, tears, breast milk and in the gastrointestinal and urogenital tract. In serum, the IgA molecule is commonly found as a monomer of either IgA1 or IgA2 subclass but can also exist as dimers or larger polymeric forms. The IgA found in secretions (secretory IgA or sIgA) is commonly a dimer with two IgA molecules joined by the J chain. Secretory IgA is the most important Ig isotype in mucous surfaces, the area of entry for most pathogens.

IgG
IgG (Immunoglobulin G) is the most abundant Ig isotype in serum and makes up approximately 80% of all serum Immunoglobulins. In humans, there are four subclasses of IgG with highest serum concentrations of IgG1 followed by IgG2, IgG3 and IgG4. In mice the IgG subclasses are defined IgG1, IgG2a, IgG2b and IgG3. The IgG molecule consists of two heavy and two lights chains (κ or λ) resulting in a molecule with two arms for antigen binding. High levels of IgG antibodies are induced following the initial IgM response in a typical immune response to antigens.
