Intended use

This monoclonal antibody enables specific detection of monkey TNF-α-producing cells in flow cytometry.

Serum/Plasma samples

Recommendation

5 µl is recommended for staining of 1 million cells in a total volume of 50 µl. PF647P is excited by the red laser (633 nm). The excitation max is 654 nm and the emission max is 672 nm.

Detection of TNF-α by flow cytometry in viable lymphocytes from Rhesus macaque. Stimulated, fixed and permeabilized cells were stained with anti-human/monkey mAb MT15B15-PF647P (left panel). Matched isotype control antibody (right panel). Flow cytometry was performed on a BD FACSVerse system.

The systems reactive with NHPs are either based on cross-reactive human kits or specifically developed monkey kits (NHP).

Cross-reactivity verification tests have been performed in ELISpot and/or ELISA by Mabtech and/or by others. Evaluations of assays with less solid evidence of cross-reactivity are shown as symbols within parentheses.

Analyte description

Tumor necrosis factor (TNF), also known as TNF-α, is produced by many different cell types, e.g., monocytes, macrophages, T cells, and B cells. Among the many effects of TNF-α are protection against bacterial infection, cell growth modulation, immune system regulation, and involvement in septic shock.
